Background The small-molecule BC-11-38 is an inhibitor of dual 3',5'-cyclic-AMP and -GMP phosphodiesterase 11 (PDE11), which catalyzes hydrolysis of the second messengers cyclic guanosine 3', 5'-monophosphate (cGMP) and cyclic AMP (cAMP) in intracellular signaling regulation. BC-11-38 is a selective and potent inhibitor of PDE11, displaying IC values of less than 330 nM for PDE11 and a greater than 100-fold selectivity for PDE11 relative to other PDE isozymes. BC-11-38 treatment of human adrenocortical cells inhibited PDE11 and resulted in increased cAMP levels, cAMP-dependent protein kinase (PKA) activity, and cortisol production (1,2).
