Background Zinc finger protein 384 (ZNF384) was first characterized as a C2H2-type zinc finger protein, functioning as a transcription factor regulating expression of extracellular matrix genes (1). ZNF384 was later shown to also regulate genes involved in cell growth and metastasis (2). ZNF384 has been predominantly studied in B-cell leukemias, particularly B-cell acute lymphoblastic leukemia (B-ALL) (3,4). In these tumors, ZNF384 has aberrant expression, resulting in dysregulation of target genes and mutations resulting in oncogenic fusion proteins (3-5). ZNF384 fusions with EP300, TCF3, TAF15, and CREBBP are the most commonly studied in these cancers (3,4,8). Recent research has focused on mutations and altered expression of ZNF384 in breast cancers (6,7). ZNF384 and its related fusion proteins are being investigated as therapeutic targets in leukemia and other cancers (8).
