Background p400, also known as EP400 or Domino, is a chromatin remodeler initially found to interact with the adenovirus oncoprotein E1A (1,2). p400 is part of the TIP60 histone acetyltransferase complex and is an ATPase capable of catalyzing the exchange of histone H2A with the variant histone H2AZ (3,4). This unique role of p400 has been shown to affect important processes by modifying gene expression through changes in chromatin structure. p400 plays a role in gene expression during embryonic hematopoiesis (5). Also, p400 can regulate p21 to control cell cycle and senescence (6,7). p400 is recruited to sites of double-stranded breaks to destabilize nucleosomes, resulting in RNF8-mediated chromatin ubiquitination and recruitment of BRCA1, 53BP1, and Rad51 (8,9). The activity of p400 is important in the development and survival of Schwann cells and oligodendrocytes (10,11). In addition to H2A/H2AZ exchange, p400 can also exchange H3 with the variant H3.3 to contribute to gene regulation (12).
