Product Usage Information Western Blotting: 1:1000 Storage Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at -20°C. Do not aliquot the antibody. Protocol Available protocols: Western Blotting Specificity / Sensitivity SNAP-Tag (F9J8I) Rabbit Monoclonal Antibody detects recombinant proteins containing the SNAP epitope tag. This antibody recognizes the SNAP-tag sequence fused to either the amino or carboxy terminus of targeted proteins in transfected cells. This antibody detects exogenously expressed SNAP or CLIP-tagged proteins in cells expressed under a CMV promoter. Expression under other promoters has not been evaluated. Species Reactivity: All Species Expected Source / Purification Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Ala62 of SNAP-tag protein. Background Epitope tags are useful for the labeling and detection of proteins using immunoblotting, immunoprecipitation, and immunostaining techniques. Tags can be genetically-encoded and expressed on the protein-of-interest, and subsequently detected using antibodies or other methods, including synthetic probes. SNAP-tag is a 20 kDa mutant of the DNA repair protein O -alkylguanine-DNA alkyltransferase (AGT) that, when "tagged" to the protein-of-interest, can be used to measure the tagged protein's expression, localization, and interactions with other proteins. AGT reacts specifically and rapidly with ectopically applied benzylguanin derivative substrates, resulting in covalent labeling of SNAP-tag (1). The benzylguanin derivatives, when labeled with fluorophores, for example, can be used as irreversible probes for SNAP-tagged fusion proteins. SNAP-tag can also be independently detected and labeled using a SNAP-tag protein-specific antibody using a variety of immuno-based assays.
