Background
The cytoskeleton consists of three types of cytosolic fibers: actin microfilaments, intermediate filaments, and microtubules. Peripherin is a class-III intermediate filament protein selectively expressed in peripheral nervous system (PNS) neurons (1). Several peripherin isoforms exist through alternative splicing, with the dominant isoform occurring at 58 kDa and minor isoforms occurring at 48, 45, 32, and 25 kDa (2,3). Additionally, an aggregate-forming 28 kDa splice isoform of peripherin has been shown to be upregulated in Amyotrophic Lateral Sclerosis, suggesting a role in disease pathology (3). Functionally, peripherin plays an important role in neurite outgrowth, axonal transport, and myelination; it also interacts with proteins responsible for vesicular trafficking (4). Because of its nearly exclusive expression in PNS neurons, it may be a promising blood biomarker of PNS axonal damage (5).
