Product Usage Information To block the interaction of IgG with mouse Fc receptors, add 1.0 μg of antibody per million cells in 100 µl of buffer. Incubate 10 mins at 4ºC or room temperature as desired. A wash step is not needed prior to adding antibodies of interest. The optimal blocking concentration may be assay dependent and may require titration. Background The 2.4G2 clone is widely used as a blocking reagent to prevent interactions between mouse Fc receptors and IgG. By binding to CD32 (FcgammaRII) and CD16 (FcgammaRIII), the 2.4G2 clone prevents the Fc receptor proteins from being able to interact with the Fc domain of antibodies subsequently added for analysis. Performing an Fc blocking step thus reduces background and false positive events in flow cytometry assays on live cells.
