This antibody specifically detects TERT isoform 1 (O14746-1) and TERT isoform 3 (O14746-3) proteins and an 85 kDa protein of unknown identity in some cell lines. Species Reactivity: Human Source / Purification Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human TERT protein. Background Telomerase reverse transcriptase (TERT) is a key component of telomerase (1), which is a ribonucleoprotein (RNP) complex responsible for extending telomere DNA to maintain telomere length and avoid cell death. TERT expression is often very low in normal cells, with the exception of those that require continuous proliferation (2). Increased telomerase is universal in human cancers (3). In cancer, TERT activity is mainly regulated by transcriptional activation through promoter hotspot mutations (4). Gene amplification through chromosomal rearrangement and promoter methylation also play roles in upregulating TERT (5). Germline reduction in telomerase precipitates tissue-failure diseases characterized by very short telomeres, such as aplastic anemia, dyskeratosis congenita, and pulmonary fibrosis (6).
