Background Zinc finger and SCAN domain-containing protein 4 (ZSCAN4) is a transcription factor that is selectively expressed in a unique subpopulation of embryonic stem (ES) cells. Embryonic development is initially controlled by maternally-deposited transcripts and subsequently by zygotic transcription, following the maternal-to-zygotic transition and zygotic genome activation (ZGA) (1). ZGA occurs at the 2-cell (2C) stage of embryonic development, and ZSCAN4 is a specific marker of this stage for embryo and ES cells (2). ZSCAN4 has been shown to regulate telomere maintenance in mouse ES cells and bind to nucleosomal microsatellite DNA repeats, which are prone to genomic instability in mouse 2C-like cells (3,4). ZSCAN4 expression has also been positively correlated with derepression of heterochromatin in mouse ES cells (5). Additionally, ZSCAN4 was shown to facilitate alterations to the chromatin landscape and promote the cancer stem cell phenotype in head and neck squamous cell carcinoma (6).
