Species Reactivity: Human, Mouse, Rat Source / Purification Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Pro990 of human MYO1D protein. Background MYO1D (myosin 1D, myosin 1d) is a class I myosin small motor protein with the ability to simultaneously bind to actin filaments and cellular membranes, coupling membrane and cytoskeletal movement. Like other family members, MYO1D contains an N-terminal motor domain for actin binding, followed by two calmodulin-binding domains (IQ domains), and a C-terminal tail homology (TH1) for interaction with membrane localized proteins (1). MYO1D controls the planar cell polarity (PCP) pathway by coupling actin filament and PCP proteins to regulate cellular orientation. Using this mechanism, the protein controls cellular left-right asymmetry and direction of cilia cell movement (2-4). MYO1D also regulates myelin membrane dynamics by interacting with myelin membrane proteins (CNP and PLP) to maintain normal morphology of oligodendrocytes (5-7). The C-terminal domain of MYO1D has been shown to bind the EGFR family kinase domain (8). This interaction is required for MYO1D anchoring of EGFR family RTKs in the membrane/cytoskeleton structure for ligand activation (9). Knockdown of MYO1D suppresses cancer growth, invasion, and downstream signaling (10).
