Background
DEAD box protein 42 (DDX42) is an ATP-dependent DEAD box RNA helicase that has key functions in splicing. DDX42 has RNA chaperone activity and associates with the SF3b complex along with DDX46, facilitating 17S U2 snRNP assembly (1-4). In oncogenic mutations, interactions between SF3B1 and DDX42/46 are greatly reduced, resulting in alternative splicing (5). DDX42 also plays a key role in the inhibition of positive-strand RNA viruses, including HIV and SARS-CoV-2 (6,7). In addition to its role in splicing, DDX42 is capable of binding G-quadruplexes, higher-order structures in the genome which can be aberrantly accumulated in cancer (8).
