Background Glutaredoxin-1 (GLRX1, GRX1), also known as thioltransferase-1 (TTase-1), is a thiol disulfide oxidoreductase that catalyzes protein deglutathionylation using glutathione as a cofactor. This small enzyme has a key role in redox homeostasis in the cell (1,2). Studies of mouse models show that GLRX1 is critical in maintaining hepatic lipid homeostasis and preventing fatty liver disease (3). In addition, fibrotic livers have reduced levels of GLRX1 and enhanced levels of protein S-glutathionylation. Furthermore, overexpression of GLRX1 suppresses protein S-glutathionylation and liver fibrosis (4).
