Background Clathrin-coated assembly protein SNAP-91, also known as AP180, is an adaptor protein that regulates the endocytosis of proteins and receptors in the membranes at synaptic vesicles (VS), and in the sorting and clustering synaptic vesicle cargo (1,2). SNAP-91/AP180 regulates the formation of clathrin lattices on lipid templates in conjunction with clathrin assembly lymphoid myeloid leukemia (CALM), and often interacts with other endocytic pathways, including the adaptor protein-complex AP-2 (3,4). SNAP-91/AP180 modulates the invagination of the clathrin-coated pit (CCP) by facilitating the clathrin assembly (5). Mutations in the C. elegans SNAP-91/AP180 neuronal homologue unc11 induces large endocytic vesicles and defective neurotransmitter processes (5,6). SNAP-91/AP180 has been associated with and is related to schizophrenia risk variants, resulting in deficits at the pre- and post-synaptic neuronal function (7). In the tau transgenic mouse model of Alzheimer's disease, Tg2576, or the overexpression of SNAP-91/AP180, a selective clathrin-dependent endocytosis of monomeric Aβ peptides results in a greater uptake of Aβ 1-42 compared to Aβ 1-40, increasing the toxicity of cells due to Aβ peptide imbalance (8,9). SNAP-91/AP180 has been proposed as a candidate biomarker for glioblastoma (GBM), along with COL3A1, where a reduction of levels of these proteins was found in patients with GBM and other malignant gliomas (10).
