Product Usage Information
This product is the carrier free version of product #46088. All data were generated using the same antibody clone in the standard formulation which contains BSA and glycerol. This formulation is ideal for use with technologies requiring specialized or custom antibody labeling, including fluorophores, metals, lanthanides, and oligonucleotides. It is not recommended for ChIP, ChIP-seq, CUT&RUN or CUT&Tag assays. If you require a carrier free formulation for chromatin profiling, please contact us . Optimal dilutions/concentrations should be determined by the end user. BSA and Azide Free antibodies are quality control tested by size exclusion chromatography (SEC) to determine antibody integrity.
Formulation

Storage
Store at -20°C. This product will freeze at -20°C so it is recommended to aliquot into single-use vials to avoid multiple freeze/thaw cycles. A slight precipitate may be present and can be dissolved by gently vortexing. This will not interfere with antibody performance.
Specificity / Sensitivity
DAP12 (E8P9U) Rabbit Monoclonal Antibody (BSA and Azide Free) recognizes endogenous levels of total DAP12 protein. Non-specific staining was observed in testis by immunohistochemistry.

Background
DNAX-activating protein 12 (DAP12, TYROBP) is a signaling adaptor for several pathogen receptors expressed by cells of the innate immune system (1). The DAP12 protein structure consists of a short extracellular domain, a transmembrane domain, and a cytoplasmic immunoreceptor tyrosine-based activation motif (ITAM) (2). DAP12 protein is expressed by hematopoietic cells, including NK cells, monocytes, macrophages, dendritic cells, mast cells, basophils, eosinophils, neutrophils, and some γδ T cells and NKT cells (1). DAP12 exists as a homodimer that associates with a variety of receptors involved in pathogen detection, including the KIR family of NK cell receptors (2,3). Ligand binding by DAP12-associated receptors results in phosphorylation of tyrosine residues within the DAP12 ITAM by Src family kinases and leads to activation of Syk or Zap-70 and downstream signaling responses (2).
