Background PUS7 is a pseudouridine synthase that can catalyze pseudouridylation of various RNA species (1-3). It is critical for the pseudouridylation of the U2 small nuclear RNA, the branch recognizing portion of the spliceosome, at position 35, which promotes pre-mRNA splicing (2). PUS7 deposits pseudouridine on both mRNAs and snoRNAs, which affects stability (4). Furthermore, during development, PUS7 pseudouridylates tRNA-derived small fragments, activating and targeting them toward the translation initiation complex (5). mutations have been described in intellectual disability and microencephaly (6,7). PUS7 has been implicated in colorectal cancer by binding and stabilizing SirT1, activating Wnt signaling (8).
