Background Membrane-bound O-acyltransferase domain-containing protein 7 (MBOAT7), also called lysophosphatidylinositol acyltransferase 1 (LPIAT1), is an enzyme involved in phospholipid metabolism by facilitating the addition of phosphatidylinositol onto arachidonic acid (1,2). MBOAT7 can promote ferroptosis, an iron-dependent form of cell death characterized by peroxidation of polyunsaturated fatty acids (3). MBOAT7 plays an important role in diseases, including liver disease, cancer, and neurodegeneration. Loss of MBOAT7 in mice results in atrophy of the cerebral cortex and hippocampus (4). MBOAT7-driven lipid remodeling promotes the progression of clear cell renal carcinoma (5). In non-small cell lung cancer, MBOAT7 levels are elevated and support prostaglandin synthesis, cell proliferation, and anchorage-independent growth (6).
