The NEBNext High-Input Poly(A) mRNA Magnetic Isolation Module is designed to isolate intact poly(A)+ RNA from high inputs (5-50 µg per 0.2 ml reaction) of previously isolated total RNA, using oligo d(T) Related Categories Next Generation Sequencing Library Preparation FAQ Q: Can I isolate poly(A) mRNA directly from tissue lysis, using the NEBNext® High Input Poly(A) mRNA Isolation Module? A: No, this kit is intended for isolation of mRNA from previously isolated total RNA. Total RNA can be isolated using the Monarch® Total RNA Miniprep Kit (NEB #T2010). Q: Do I need to perform two rounds of binding? A: Multiple RNA binding events are recommended to obtain high specificity for poly(A) RNA and minimize background. Q: Can I use the NEBNext® High Input Poly(A) mRNA Isolation Module with total RNA inputs higher than 50 µg? What about lower inputs? A: For best results when using total RNA inputs higher than 50 µg, all volumes in the protocol can be proportionally scaled. For inputs less than 5 µg, the NEBNext® Poly(A) mRNA Magnetic Isolation Module (NEB #E7490) is recommended. Q: Can the RNA be eluted with water? A: Yes, the RNA can be eluted with water. The elution volume can also be adjusted for a higher or lower final concentration.
