A: HinP1I cleaves to leave a different overhang. HinP1I preoduces a 5´ extension, whereas HhaI produces a 3´ extension. The 5´ extension can be efficiently ligated into the AccI site of M13 and pUC cloning vectors. Both are blocked by CpG methylation. Q: Does HinPI cleave single-stranded DNA? A: Yes, at about half its rate on double-stranded DNA. It is unclear if the enzyme cuts single stranded DNA or at double stranded regions in single stranded DNA. Q: What is the activity of Hinp1I at 25°C? A: HinPI is approximately 25% active at 25°C versus 37°C. Q: Do I have to set-up digests with Time-Saver™ qualified enzymes for 5-15 minutes? Can I digest longer? A: NEB's Time-Saver™ enzymes have the benefit of working fast (5-15 minutes), but are also designed and qualified to withstand overnight digestions without degradation of DNA. Q: Is this enzyme sensitive to dam, dcm or mammalian CpG methylation? A: Yes. This enzyme cannot cut recognition sites in mammalian gDNA that are blocked by CpG methylation. We are also in the process of moving all restriction enzyme formulations to contain rAlbumin. We feel that moving away from animal-containing products is a step in the right direction and are able to offer this enhancement at the same price.
