Overview
The HTRF Mutant Ataxin 2 standard has been validated for use with the HTRF Mutant Ataxin 2 kit, but is not included in the kit, and must be purchased separately.
The standard is supplied as a frozen stock solution at a concentration of 75,000 pg/mL. When following the recommended protocol for diluting the standard the HTRF Human Mutant Ataxin-2 assay has a detection range of 78 – 15,000 pg/mL, a limit of detection (LOD) of 10 pg/mL, and a limit of quantitation (LOQ) of 49 pg/mL.

Using the HTRF Human Mutant Ataxin 2 Standard with the HTRF Mutant Ataxin 2 Kit
The HTRF Mutant Ataxin 2 standard was developed to be used in conjunction with the HTRF Human Mutant Ataxin 2 Detection Kit to quantify the amount of mutant Ataxin 2 present in cell lysate samples.  The HTRF Mutant Ataxin 2 assay is based on a TR-FRET sandwich immunoassay involving two specific antibodies, one labelled with Eu3+-cryptate (donor) and the other with d2 (acceptor). One antibody is directed against the total Ataxin 2 protein, and the second recognizes specifically the mutant polyQ domain. Both antibodies bind to soluble Mutant Ataxin 2, and the donor-acceptor proximity leads to a fluorescent TR-FRET signal. The intensity of the signal is directly proportional to the concentration of soluble Mutant Ataxin 2 present in the sample (cell lysate or tissue lysate).

Protocol for using the HTRF Human Mutant Ataxin 2 Standard
The HTRF Human Mutant Ataxin 2 Standard is supplied as a frozen stock solution at 75,000 pg/mL.  This stock solution is diluted in HTRF Lysis Buffer 1 to create a standard curve ranging in concentration from 0 – 15,000 pg/mL.  The HTRF Mutant Ataxin 2 assay can be run in a 96- or 384-well low volume white detection plate (20 µL final). As described here, samples (cell/tissue lysates) or standards are dispensed directly into the assay plate for the detection of Mutant Ataxin 2 by HTRF reagents. The antibodies labelled with HTRF fluorophores may be pre-mixed and added in a single dispensing step. No washing steps are needed.
