Overview
TAR DNA binding protein 43 (TDP-43) is a DNA and RNA-binding protein which plays a crucial role in RNA metabolism. Mainly located in the nucleus, TDP-43 shuttles between the nucleus and the cytoplasm. In pathological conditions, including stress or mutation, TDP-43 is mislocalized in the cytoplasm, which can be cleaved in C terminal fragments, thus leading to an accumulation of ubiquitinated and hyperphosphorylated toxic aggregates and then insoluble inclusion bodies. Aggregated TDP-43 has been identified as a hallmark of amyotrophic lateral sclerosis (ALS) and frontotemporal lobar dementia (FTLD), and more widely in several neurodegenerative diseases such as Alzheimer’s disease.
The AlphaLISA SureFire Human and Mouse Total Aggregate TDP-43 Detection Kit is a sandwich immunoassay for the quantitative detection of total aggregate TDP-43 in cellular lysates, using Alpha Technology.
